Was that a threat so I couldn’t resign?

I'm currently employed at a bank here in the Philippines but I already expressed my intent to resign. I submitted my resignation last February 14 with a 30-day notice, thus completing the 30-day notice period on March 16. My supervisor already signed and acknowledged my resignation letter and I am about to prepare the necessary documents/clearance for me to properly quit my job. By the way, I've also been accepted at a different job which I will be starting on March 21. The thing now is, I got to talk to my supervisor and she said she can't proceed with my resignation since I haven't turned over my tasks/responsibilities. I couldn't do the turn over since they haven't assigned anyone to replace me. She insisted that I extend my resignation on the 31st which makes it a conflict to my soon-to-be job. My question now is, what should I do?…
