ADA protection in the workplace?

I have been at my job for one year. From offer negotiation, I've had three conversations with my manager about WFH two days a week as I have Autism and am most comfortable not processing external stimuli 5 days a week. He's denied me everytime saying I need to be flexible and he's “unwilling to accommodate” me. My male coworker gets to WFH 4 days a week while his wife is on bedrest during pregnancy. My boss is a typical micromanaging narcissist and I'm burnt out. I'm ready to leave without a new job lined up (I'm actively interviewing, it is just too daunting with a huge workload) but want to understand if I would have a case for unemployment based on discrimination as protected under the ADA. Any help is much appreciated.
