Is this a threat?

Long story short: I got into a standoff with my manager today. I work full time, Monday to Friday. I asked her if I could work 7-3 or 6-2 (as I originally was scheduled at that time from the previous manager), but she scheduled me for 9-5 for the past month. I said that I can no longer work this hours due to commitments. I have to pick my sister up from school sometimes at 4pm, and it takes me 45 minutes to get there from my work. She said that she can’t accommodate at all, and that I am expected to work any shift I am assigned as I am a full time worker. She said “you either work the hours you’re assigned, or you don’t work for us at all”. Is this a threat? Should I raise this to her boss?
