Saying no to taking on an additional role or responsibilities

Have you ever said “no, but thank you” to being approached and asked to take on a new or additional role and/or responsibilities? For example, something that takes time away from what you were hired to do/like to do and which may not be as marketable for the next job. Unless you want to be pigeonholed into endless grunt work and you are too far along into your career to do that. More doesn't mean better. If so, how did that turn out? Any tips on how to successfully navigate “no, but thanks”.  The question is the same if there is significantly more workload but no extra salary, or, an offer of a promotion that you don't actually want (title and more responsibility but not much extra pay to offset).  Or is “no” simply something that no one ever says. ​ Thank you.
