Asking me for resignation letter to get PTO

So, I work for a research project at a reputable university, I was informed that the end of my contract is coming at the end of November although I was expecting to stay until Spring of next year. My supervisor told the that the HR person recommended that I should submit a resignation letter 4 weeks before the end of the contract to make sure I get paid for the PTO I have earned, is this normal? I’m not sure I qualify for unemployment insurance in this case but I’m pretty sure that by submitting a resignation letter would definitely make me ineligible. Would appreciate any information on how to handle this. Thanks!
